Systems Biology: Networks @ CSH 雑感
Cold Spring Harbor Laboratory のミーティングシリーズの一つ、Systems Biology: Networksに行ってきました。基本的にClosedなミーティングなので、ここでは、論文として公開されている情報にポインタが張れるレベルの内容を記載します。(興味がある方は、私が理解した&覚えている範囲でお伝えしますので、個人的に連絡ください。CSHのミーティングでは、personal communicationでの伝達は認められています)
私自身は主として生命科学に応用を持つアルゴリズムやソフトウエアの開発が主な研究対象なので、Systems Biologyを専門にしているわけではありません。そういう意味で、今回のミーティングは、私の研究にぴったり合っている内容ではありませんでした。それでも、自分が開発した手法がどのように活かせるか、どのような手法を開発するのが良いか、世の中の生物学がどのような方向に進んでいくのかを知るのに、良い機会なので、会議に出て勉強するようにするのが、今回の目的でした。
- 自分が読んでいた論文のFirst authorがPIに準じる立場になって論文を出してきている
世代が新しくなっているという事と同義です。Systems Biologyの基となるデータを作り、解析した人たちがPIになって次の世代を作り上げて行っている感じがあります。片手に収まる範囲の年数で結果を出してきているので、周辺の巨匠も手厚くサポートをしていることと思います。
- 会議オーガナイザーが関連した研究発表が多い
今回の発表では、このミーティングのオーガナイザーが関連した研究が、Oral talkに多く選ばれている印象がありました。それも、どの研究もレベルが高いので、故意に選んだというより、Winner takes all に近い状況が生まれつつあるのだと思います。
- 題材選びの重要性
上記に関連していますが、今は対象をうまく取った研究室が勝っている印象があります。手法として新しいものを開発しているのではなく、分子生物学に多く見られる様に、既存の手法を美味く組み合わせて生物学的問題を解く事が多く行われている印象が強いです。対象となる遺伝子やパスウエイも、古来の分子生物学の様な1遺伝子よりは多いですが、データそのものは網羅的に採取しつつも、もともと対象とする現象やパスウエイが存在していて研究をスタートしている研究室が成功している雰囲気でした。
- 情報科学的なアプローチは孤立し、生物学的なアプローチの補助に廻ったものが成功している
更に上記との関連になりますが、会議全体として、「生物学」でした。大規模データから網羅的に遺伝子・タンパク質を取得しても、結局は興味有る(差異が出た)現象に着目して、古来の生物学に落とし込んで勝負しています。結果として、生物学が主、情報科学が従の印象が強いです。情報科学的なアプローチを取っているグループもあるようでしたが、全体の反応はいまいちに見えました。(個人的には、情報科学的なアプローチを開発、多様して生物に迫りたいのですが)
- 新しい手法が開発されているわけではない
次世代シークエンサの発達もあり、画期的なアプローチが開発されているのかとも思ったのですが、幾つか次世代シークエンサを利用した発表があった(illuminaを利用したタグの取得、Y2Hの高速化・低価格化 (Yu et al. Science 2008 http://www.sciencemag.org/cgi/content/abstract/sci;297/5588/1837 ))だけで、既存手法を広範囲で適用した話が多い印象。私が知らなかったのは、Yeast one-hybridとそれに関連した手法でした。いくつかの研究室で使われている所を見ると、私が無知なだけで、割と一般に使われそうですね。
- でもネタは全て新しい
発表内容のほとんどが2008年以降の論文発表を基にしたもので、参照論文も2005年以前のものは、ほとんど無い状態。
- データの精度について考えて居る発表が散見された
多くの生物学的なデータは統計的有意性を調べてp-valueを計算して「ほーら、違いがあるでしょ」というパラダイムが多いわけですが、Systems biologyで扱われるデータは多変量の為に、既存の手法ではp-valueが出せない場合が多く現れます。そのような場合のデータの精度について、議論している人が何人か居ました。http://www.nature.com/nbt/journal/v23/n7/abs/nbt1116.html など。(統計屋さんんが居たら嫌うような、結構乱暴な仮定や、いい加減な議論も見られました。)
- 雑誌のCELLがアグレッシブ
CELLの論文が良く参照されていた気がしたのですが、帰ってきてCELLを眺めてみると、昔のpure biologyな非常に格調高く、お堅い印象な論文だけでなく、in silicoの論文も最近は積極的に採用している様な雰囲気に見えました。チャンス到来か!?

- 論文じゃないけど:アイディアに、頭の良い(スマートな)学生やポスドクを投入すると、すばらしいアイディアになり、そこに幸運と努力(汗)が加わると、NIH/NSFのファンディングに繋がり、結果、頭の良い学生やポスドクが得られて、投入できる。という正のフィードバックがおもしろかった。
